X-linked severe combined immunodeficiency disease and the gamma c receptor component: prospects for molecular diagnosis.

چکیده
The role of interleukin-2 (IL-2) in immune function has been studied extensively. Cellular receptors for IL-2 (IL-2R) on lymphocytes and certain other cells have been characterized and are responsible for transmitting the cellular response to IL-2. The importance of the IL-2R " chain has been emphasized by analyses of IL-2R expression on functional T cells and natural killer (NK) cells and by its role in the pathophysiology of X-linked severe combined immunodeficiency disease (XSCID). This minireview summarizes these findings, emphasizing the complexities in establishing a clinical assay system for characterizing defects in the IL-2R " subunit and the atypic expression of these defects in patients with SCID or their unaffected carriers.

Shared gamma(c) subunit within the human interleukin-7 receptor complex. A molecular basis for the pathogenesis of X-linked severe combined immunodeficiency.
Genetic evidence suggests that mutations in the gamma(c) receptor subunit cause X-linked severe combined immunodeficiency (X-SCID). The gamma(c) subunit can be employed in receptor complexes for IL-2, -4, -7, -9, and -15, and the multiple signaling defects that would result from a defective gamma(c) chain in these receptors are proposed to cause the severe phenotype of X-SCID patients. Interest...
متن کاملMutation analysis of IL2RG in human X-linked severe combined immunodeficiency.
Severe combined immunodeficiency (SCID) is a syndrome of profoundly impaired cellular and humoral immunity. In humans, SCID is most commonly caused by mutations in the X-linked gene IL2RG, which encodes the common gamma chain, gamma c, of the leukocyte receptors for interleukin-2 and multiple other cytokines. To investigate the frequency and variety of IL2RG mutations that cause SCID, we analyz...
متن کاملRestoration of lymphoid populations in a murine model of X-linked severe combined immunodeficiency by a gene-therapy approach.
X-linked severe combined immunodeficiency (XSCID) is a life-threatening syndrome in which both cellular and humoral immunity are profoundly compromised. This disease results from mutations in the IL2RG gene, which encodes the common cytokine receptor gamma chain, gamma(c). Previously, we generated gamma(c)-deficient mice as a murine model of XSCID. We have now used lethally irradiated gamma(c)-...
متن کاملA mutation in the IL-2 receptor gamma chain gene associated with X-linked severe combined immunodeficiency accompanying opisthotonus.
Severe combined immunodeficiency (SCID) is an inherited disease with profoundly defective T cells, B cells, and NK cells. X-linked severe combined immunodeficiency (X-SCID) is its most common form. In this report, we describe a 4-month old male with X-SCID who also showed opisthotonic posturing. Opisthotonus represents abnormal motor posturing and is defined as the posturing, in which the neck ...
متن کاملA New IL-2RG Gene Mutation in an X-linked SCID Identified through TREC/KREC Screening: a Case Report.
Severe combined immunodeficiency (SCID) represents a rare group of primary immunodeficiency disorders (PIDs), with known or unknown genetic alterations. Here, we report a new interleukin 2 receptor, gamma chain (IL-2RG) mutation in an Iranian SCID newborn. The patient was a 6-day old boy with a family history of PID. The child was screened using a molecular-based analysis for the assessment of ...
